MGMT (human) is located at chromosome 10 (10q26). Encoding the DNA repair protein O-Methylguanine-DNA Methyltransferase. MGNT repairs alkylated guanine in DNA by stoichiometrically transferring the alkyl group at the O-6 position to a cysteine residue in the enzyme. Frequently loss of gene expression occurs via hypermethylation of CpG sites within the promoter region of the MGMT gene.

Short Description Pyrosequencing is the method of choice if you look for polymorphisms (point mutations on different allels), whereby even the most difficult SNPs and mutations can be analysed.
Pyrosequencing gives unambiguous genotyping results as well as some redundant sequence information beyond the SNP position, which serves as a valuable internal control, obtained for each sample. Pyrosequencing presents SNPs in the context of the surrounding sequence. This guarantees that each individual analysis is correct, without the need for other controls.
